though i like using phase and echo, if forced to only one pedal, i would use my JHS Morning Glory. it just makes any amp/guitar combination sound so sweet.

EHX Tone Tattoo. Bit of a cheat, 'cos it's a Memory Toy, Neo Clone and Metal Muff all in one case, plus a noise gate for good measure!

Probably either a wah pedal or delay/vibrato/flanger pedal. My Peavey 6505 amp has tons of power and distortion and after playing it with my Gibson Flying V, no need for much else. Fortunately such a pedal exists:

I have tons of effects on my cheap Vox Mini G3 amp that I bought for taking camping and to the beach since it is tiny has great sound and uses batteries. For a single pedal, I would have to say a looper! Being able to loop chord backing tracks while playing guitar solos is so important when playing alone. My guitar has plenty of kick already and the amp does a nice job on distortion.
